3D cell culture using gas permeable membrane

Gas Permeable VECELL® G-Plate

G-Plate scaffold consists of a unique 3D cell culture model by high oxygen permeable materials and application of artificial vessel techniques for fabrication. The features of transparency and surface smoothness prevent diffuse reflection and spreading of light source, and are suitable for imaging system including microscope observation from the bottom and drug discovery screening. Unlike spheroids, there is no cell death towards the center of the spheroid due to unique form and oxygen supply. A high Z-factor supports analysis of even the inside of the 3D structure. Cell sizes are close to natural tissue and nuclei are also compact.

Cell culturing by conventional plastic plate makes shortage of oxygen at bottom.

Most cells on VECELL® G-Plate were alive, while those on plastic plate were dead for both 96 well plate and 384 well plate.

Application

HCA (High Content Analysis) and HCS (High Content Screening) can analyze the effects of compounds or target substances to a sample of cells quickly and with detail. Therefore, these methods are becoming important in drug development screening. The analysis should be carried out under the same situation as in vivo for cells to retain natural configuration. However, cells extend two-dimensionally in general plastic plates. On the other hand, cells which are cultured in oxygen permeable VECELL plates are suitable for HCA and HCS with ample supply of oxygen from the bottom of the plate to allow a monodispersity state without aggregation. Therefore, VECELL plates are perfect for HCA/HCS.

Cultured layers like monolayer culture are constructed and form tissue which produces ECM. Culturing with VECELL enables sectioning to easily observe growth of cells and ECM.
