Anti GPNMB, ITIH4, WDR45L, TSPO, CD163, PRDX

ITIH4(Inter-alpha Trypsin Inhibitor Heavy Chain H4)

Background

ITIH4 is the inter-alpha-trypsin inhibitor heavy chain family, member 4. The protein encoded by this gene is secreted into the blood, where it is cleaved by plasma kallikrein into two smaller forms. Expression of this gene has been detected only in liver.

GPNMB(Glycoprotein Nonmetastatic Melanoma Protein B)

Background

The transmembrane glycoprotein NMB (GPNMB) is a type I transmembrane glycoprotein which shows homology to the pmel17 precursor, a melanocyte-specific protein. GPNMB has been reported to be expressed in various cell types, including melanocytes, osteoclasts, osteoblasts, dendritic cells and to be overexpressed in various cancer types.

Untransfected (red) and transfected HeLa cells with ICAFectin441 and plasmid encoding human GPNMB (blue) were stained with Rabbit anti-human GPNMB Polyclonal IgG Antibodies (diluted 1/100) and R-PE goat anti rabbit IgG secondary antibody.

WDR45L(WD Repeat-containing Protein 45-like)

Background
WDR45L is an ubiquitously expressed WD40 repeat protein and is upregulated in a variety of tumor tissues including ovarian and uterine cancers. WD40 repeats are found in a number of eukaryotic proteins that coordinate multi-protein complex assemblies. WD40 proteins are implicated in many functions including adaptor/regulatory modules in signal transduction, pre-mRNA processing and cytoskeleton assembly.

Immunohistochemistry:
Hela cells transfected with plasmid encoding mouse WDR45L were fixed, permeabilized and stained with Rabbit anti- mouse WDR45L Polyclonal Antibodies diluted 1/100 and Alexa488 goat anti mouse IgG secondary antibody. Nuclei were counterstained with DAPI.
ELISA:
Lysates of untransfected and transfected HeLa cells with plasmid encoding mouse WDR45L were coated in 96 wells plate. Determination anti- mouse WDR45L polyclonal IgG in the mouse sera was performed by using HRP goat anti mouse IgG secondary antibody.

IAnti mouse TSPO [Translocator protein ]

Background

Translocator protein (TSPO) is an 18 kDa protein mainly found on the outer mitochondrial membrane. It interacts with StAR (steroidogenic acute regulatory protein) to transport cholesterol into mitochondria. It was first described as peripheral benzodiazepine receptor (PBR), a secondary binding site for diazepam, but subsequent research has found the receptor to be expressed throughout the body and brain. In humans, the translocator protein is encoded by the TSPO gene. It belongs to family of tryptophan-rich sensory proteins.

Flow cytometry:
Untransfected (red) and transfected with ICAFectin® 441 and plasmid encoding mouse TSPO Hela cells were fixed, 24h post transfection, permeabilized and stained with rabbit anti-mouse TSPO polyclonal antibodies diluted at 1/100 (orange) and at 1/200 (blue) and R-PE goat anti rabbit IgG secondary antibody at the dilution 1/200.

Hela cells transfected with ICAFectin® 441 and plasmid encoding mouse TSPO were fixed 24h post transfection, permeabilized and stained with rabbit anti-mouse TSPO Polyclonal Antibodies diluted 1/20 and Alexa488 goat anti rabbit IgG secondary antibody at dilution 1/1000. Nuclei were counterstained with DAPI in blue.

 

Anti mouse CD163 [Cluster of Differentiation 163]

Background
CD163 is a scavenger receptor for the hemoglobin-haptoglobin complex. It has also been shown to mark cells of monocyte/macrophage lineage. The receptor belongs to the scavenger receptor cysteine rich family type B and consists of an 1048 amino acid residues extracellular domain, a single transmembrane segment and a cytoplasmic tail with several splice variants.

Immunohistochemistry:
Hela cells transfected with ICAFectin® 441 and plasmid encoding mouse CD163 were fixed 24h post transfection, permeabilized and stained with rabbit anti-mouse CD163 Polyclonal Antibodies diluted 1/20 and Alexa488 goat anti rabbit IgG secondary antibody at dilution 1/1000. Nuclei were counterstained with DAPI in blue.
Flow cytometry:
Untransfected (red) and transfected with ICAFectin® 441 and plasmid encoding mouse CD163 Hela cells were stained with rabbit anti-mouse CD163 polyclonal antibodies diluted 1/100 (orange) and 1/200 (blue) and R-PE goat anti rabbit IgG secondary antibody at the dilution 1/200.

Anti mouse PRDX4 [Peroxiredoxin-4]

Background

Peroxiredoxin-4 is an antioxidant enzyme and belongs to the peroxiredoxin family. The protein is localized to the cytoplasm. Peroxidases of the peroxiredoxin family reduce hydrogen peroxide and alkyl hydroperoxides to water and alcohol with the use of reducing equivalents derived from thiol-containing donor molecules. This protein has been found to play a regulatory role in the activation of the transcription factor NF-kappaB.

Hela cells transfected with ICAFectin441® and plasmid encoding mouse PRDX4 were fixed 24h post transfection, permeabilized and stained with mouse anti-mouse PRDX4 Polyclonal Antibodies diluted 1/20 and Alexa488 goat anti mouse IgG secondary antibody at dilution 1/1000. Nuclei were counterstained with DAPI in blue.
Flow cytometry:
Untransfected (black) and transfected with ICAFectin® 441 and plasmid encoding mouse PRDX4 (red) Hela cells were fixed 24 hours post transfection, permeabilized and stained with mouse anti-mouse PRDX4 Polyclonal Antibodies diluted 1/100 and R-PE goat anti mouse IgG secondary antibody at the diltion 1/200.
ELISA:
Lysates of untransfected and transfected HeLa cells with ICAFectin® 441 and plasmid encoding mouse PRDX4 were coated on MaxiSorp plates (Nunc) in bicarbonate buffer. Determination of anti-mouse PRDX4 Polyclonal IgG in the mouse sera was performed by using HRP goat anti mouse IgG secondary antibody at the dlition 1/5000.
