Anti CS [Chondroitin Sulfate] (1B5)

Background

Monoclonal antibody 1B5 recognises unsulfated unsaturated disaccharide neoepitopes (i.e. C-0-S "stubs") generated at the non-reducing terminal of Chondroitin Sulfate glycosaminoglycan chains that have been pre-digested with either Chondroitinase ABC or Chondroitinase ACII [see Figure 2; Caterson B (2012) Int. J. Exp. Pathol. 93: 1 - 10].

FAQ

Q: Seeing some images of western blots (from Rees et al Biochem Journal 2000) for chondroitinase-treated tendon samples probed with mAbs 1-B-5, 3-B-3 and 2-B-6,  each antibody seems to detect a different subpopulation of digested CSPGs. However, none of the lanes appear to be controls in the form of identical samples that were not treated with chondroitinase. I was wondering if it is known to what extent these antibodies react with undigested CSPG samples; would an undigested sample run under these conditions yield a "blank" lane or are the epitopes for these antibodies present in undigested samples? If so, can you give me a sense of how the background binding would compare to the signal shown in these blots?


A: An appropriate & valid “Control” for using mAbs 1B5, 2B6 & 3B3 is a no-chondroitinase treated sample. For mAbs 1B5 & 2B6 this should give a “blank” no-binding/no colour/no fluorescence result in Western Blots and IHC of sections. However, an exception is the use of mAb 3B3 [Please see Caterson (2012) published reference]. With this mAb 3B3, its use without chondroitinase treatment [denoted by us as 3B3(-)], can give a positive result when no chondroitinase treatment is used because it recognises an unusual non-reducing terminal epitope [i.e. not a chondroitinase-generated neoepitope that is designated by us as 3B3(+)] that consists of a terminal saturated glucuronic acid residue adjacent to a 6-sulphated N-acetyl-galactosamine residue in the native CS GAG chains [see Figure 2 in Caterson (2012)]. This reaction of the 3B3(-) mAb is seen when the mAb is used for both Western Blots & IHC sections. Examples of this use of 3B3(-) & 3B3(+) are provided in Caterson (2012) attached. Interestingly, expression of the 3B3(-) epitope is a biomarker for the presence of stem/progenitor cells in many tissues.
